α-Synuclein Conformational Strains as Drivers of Phenotypic Heterogeneity in Neurodegenerative Diseases
2023, Journal of Molecular BiologyMonitoring α-synuclein aggregation
2023, Neurobiology of DiseaseCitation Excerpt :Synucleinopathies, such as Parkinson's disease (PD), dementia with Lewy Bodies (DLB), and multiple system atrophy (MSA), are a group of neurodegenerative disorders of unknown aetiology with very diverse clinical and pathological presentations. However, these diseases share certain standard features, the most significant being the aggregation of alpha-synuclein (α-syn), present in cytoplasmic inclusions bodies in the cells of affected brain regions (Papp and Lantos, 1992; Spillantini et al., 1997). These cytoplasmic inclusion bodies are diverse in nature and cellular localization across these synucleinopathies.
Autophagy in Lewy body diseases and multiple system atrophy
2022, Autophagy Dysfunction in Alzheimer's Disease and DementiaMSA: From basic mechanisms to experimental therapeutics
2020, Parkinsonism and Related DisordersCitation Excerpt :Macroscopically MSA is characterized by the abnormal accumulation of α-syn in the cytoplasm of oligodendrocytes, in the so-called glial cytoplasmic inclusions (GCIs), thus differing from Parkinson's disease (PD) where α-syn mainly accumulates in neurons in Lewy bodies [30–33]. In MSA patients, α-syn aggregates can also be found in neurons, both in cytoplasmic and nuclear inclusions [34–37]. However, the fact that subregional GCI densities correlate with the severity and the distribution of neurodegeneration supports the hypothesis that MSA is a primary oligodendrogliopathy rather than a primary neuronal pathology [38–42].
