Organization and nucleotide sequences of the human tyrosinase gene and a truncated tyrosinase-related segment

Abstract

We have isolated and sequenced the gene encoding human tyrosinase, the key enzyme in pigment biosynthesis. The human tyrosinase gene contains five exons and spans more than 50 kb of DNA on chromosome segment 11q14→q21. We have also isolated a second segment in the human genome that is closely related to tyrosinase. The tyrosinase-related segment, located on 11p11.2→cen, contains only exons 4 and 5 plus adjacent noncoding regions. This segment is present in all human ethnic groups analyzed, and the noncoding nucleotide sequences shared by the 11q tyrosinase gene and the 11p tyrosinase-related segment differ by only 2.6%. This suggests that this segment of the tyrosinase gene was duplicated approximately 24 million years ago.

      The bulky, monophenolase-blocking phenylalanine (F273 in cgAUS1) is present in most plant PPOs, but in some species leucine is at this position instead of phenylalanine, such as pineapple PPO from Ananas comosus and bread wheat PPO from Triticum aestivum [20]. However, these structurally relevant residues are not found in human and fungal tyrosinases as in Agaricus bisporus or Neurospora crassa and the fungal catechol oxidase from Aspergillus oryzae, but a thioether-forming cysteine is found in fungal tyrosinases at a different sequence location than in plant PPOs (as CXH in abPPO4 and abPPO3; and C(X)16-19H in plant PPOs, see Fig. 2) [8,20–30]. Recently, aurone synthase from C. grandiflora (cgAUS1) was successfully heterologously expressed in Escherichia coli, but recombinant aureusidin synthase (AmAS1) has not been described so far [5,8].

      In the present study, the calculated molecular mass of the AmphiTYR were 60.9 kDa. It is close to the mass (60.3–61.2 kDa) of vertebrate tyrosinase including human, chicken, mouse, cattle and fish [14,31–34], but different from the invertebrate phenoloxidase cloned from shrimps, mud crab and freshwater crayfish with 76–80.7 kDa [35–37]. Sequence data suggested that AmphiTYR had higher similarity with vertebrate tyrosinase than to invertebrate phenoloxidase.
