For example, the growth factor mediated induction of Cyr61 mRNA expression in immortalized hippocampal neuronal cells could be mimicked by activation of Raf-1 and blocked with an inhibitor of MAPK signaling [8]. Recently it has been shown that Cyr61 expression can be induced by treatment of mesenchymal stem cells with Wnt3A [9].

Furthermore, cyr61 gene expression increased significantly after stimulation with either 200 μm glutamate or 200 μm NMDA. As a positive control, cells were treated with basic fibroblast growth factor (10 ng/ml), which resulted in cyr61 induction, as previously reported (8). In a similar way, Northern blot analysis using the total RNAs isolated after being stimulated with the same concentration of etoposide, NMDA, and glutamate confirmed that IEG cyr61 is rapidly induced by these toxic stimuli (Fig. 1B).

Recent studies show that the expression of CYR61 is activated during the basic fibroblast growth factor-induced differentiation process in the embryonic hippocampal neuronal cell line H19–7 in a MAP kinase-dependent and -independent manner (61). In addition to PKC and Ca2+, muscarinic m1 receptors are known to activate MAP kinase signaling (62, 63) strongly suggesting that MAP kinase signaling may couple muscarinic receptor stimulation toCYR61 expression.
