Expression of immediate early gene cyr61 during the differentiation of immortalized embryonic hippocampal neuronal cells

Abstract

Growth factor-mediated signal transduction is a process that is of fundamental importance in understanding cellular growth and differentiation. In order to elucidate the signaling pathways leading to neuronal differentiation, we have tried to identify intermediates that are selectively induced in the differentiation of immortalized neuronal hippocampal cell line H19-7. In the present study we found that immediate early gene cyr61 is expressed in a rapid and transient manner by bFGF during the differentiation of H19-7 cells. To clarify the signal transduction pathway for the induction of cyr61 by bFGF, we checked whether Raf-1 and mitogen-activated protein kinase (MAPK) is activated during the induction of cyr61. It is identified that cyr61 is induced by bFGF via at least two signaling pathways; MAPK-dependent as well as MAPK-independent signaling pathways. This study suggested that cyr61 is likely to play an important role in neuronal differentiation process.

    The specific molecular mechanisms underlying the regulation of Cyr61 expression are in many cases unknown, but signaling pathways that have been implicated include Rho and phosphatidyl inositol 3-kinase; as well as MAPK cascades. For example, the growth factor mediated induction of Cyr61 mRNA expression in immortalized hippocampal neuronal cells could be mimicked by activation of Raf-1 and blocked with an inhibitor of MAPK signaling [8]. Recently it has been shown that Cyr61 expression can be induced by treatment of mesenchymal stem cells with Wnt3A [9].

    As shown in Fig.1A, RT-PCR analysis clearly showed that the expression of a 361-bp fragment of cyr61 was abundantly amplified after being stimulated with 85 μmetoposide for 1 h. Furthermore, cyr61 gene expression increased significantly after stimulation with either 200 μm glutamate or 200 μm NMDA. As a positive control, cells were treated with basic fibroblast growth factor (10 ng/ml), which resulted in cyr61 induction, as previously reported (8). In a similar way, Northern blot analysis using the total RNAs isolated after being stimulated with the same concentration of etoposide, NMDA, and glutamate confirmed that IEG cyr61 is rapidly induced by these toxic stimuli (Fig. 1B).

    These signal transduction mediators independently increased cellular levels of h-CYR61 mRNA, but our data do not exclude the possibility of cross-talk among them. Recent studies show that the expression of CYR61 is activated during the basic fibroblast growth factor-induced differentiation process in the embryonic hippocampal neuronal cell line H19–7 in a MAP kinase-dependent and -independent manner (61). In addition to PKC and Ca2+, muscarinic m1 receptors are known to activate MAP kinase signaling (62, 63) strongly suggesting that MAP kinase signaling may couple muscarinic receptor stimulation toCYR61 expression.
